Luca Pinello is a computational biologist and associate professor at MGH/Harvard who blends machine learning, single-cell genomics, CRISPR genome editing, and generative AI to tackle gene regulation and disease. He leads a lab that produces widely used open-source tools (CRISPResso2, CRISPRme, PrimeDesign, BEAN) and novel methods for single-cell trajectory and multimodal analysis (STREAM, Dictys, SIMBA), and his work has informed regulatory discussions around CRISPR therapies. As an associate member of the Broad Institute’s Gene Regulatory Observatory and an affiliate of the Kempner Institute, he directs large-scale functional genomics efforts within the NIH IGVF Consortium, applying base editing to dissect variant effects in heart and blood disease. His recent research extends to DNA-Diffusion, a diffusion-model approach for designing cell-type-specific regulatory sequences with therapeutic potential, reflecting a rare combination of rigorous mathematics, software engineering, and experimental collaboration.
